2'-Deoxy-4'-thioadenosine is a purine nucleoside that is synthesized from 4-thiouridine and phosphorylase. It is cytotoxic and has significant cytotoxicity against cultured cells. The cytotoxicity of 2'-deoxy-4'-thioadenosine was found to be due to its conversion to the triphosphate form in the cells, which inhibits both DNA synthesis and RNA synthesis. This drug also has an inhibitory effect on the growth of tumor cells by inhibiting ribosomal protein synthesis.
