Calciumfolinat, auch bekannt als Leucovorin oder Citrovorum-Faktor, ist ein Calciumsalz der Folsäure, das ein Derivat der Folsäure ist. Leucovorin is a folate analog drug that is used to treat the toxic effects of methotrexate and other antagonists of folate. It works by counteracting the toxic effects of methrotrexate by inhibiting the enzyme dihydrofolate reductase. It is used to increase the levels of folic acid and by strengthening the link between the active metabolite (5-FdUMP) and the enzyme thymidylate synthetase, leucovorin increases the action of fluorouracil. Additionally, in order to increase survival in patients undergoing palliative treatment for colorectal cancer, it is used in conjunction with 5-fluorouracil.
