Rimantadinhydrochlorid ist ein antivirales Medikament, das hauptsächlich zur Prävention und Behandlung von Influenza-A-Virusinfektionen eingesetzt wird. Es wird als Adamantan-Derivat klassifiziert, das wirkt, indem es den viralen Replikationsprozess hemmt. Die Substanz erscheint als weißes bis off-white kristallines Pulver und ist in Wasser löslich, was sie für die orale Verabreichung geeignet macht. Rimantadinhydrochlorid hat ein relativ niedriges Molekulargewicht und weist einen moderaten pKa auf, was auf seine schwache Basizität hinweist. Sein Wirkmechanismus besteht darin, den M2-Ionenkanal des Influenza-Virus zu blockieren, wodurch das Uncoating des viralen Partikels und die anschließende Freisetzung von viralem RNA in die Wirtszelle verhindert werden. Das Medikament wird im Allgemeinen gut vertragen, kann jedoch Nebenwirkungen wie gastrointestinale Störungen, Schwindel und Schlaflosigkeit verursachen. Aufgrund des Auftretens resistenter Stämme des Influenza-Virus war die Wirksamkeit von Rimantadin in den letzten Jahren eingeschränkt, was zur Entwicklung alternativer antiviraler Mittel führte.
