AMG 331 is a drug that belongs to the class of biguanides. It is used for the treatment of type 2 diabetes and hyperinsulinemia. Inhibition of gluconeogenesis and insulin resistance are thought to be responsible for AMG 331’s therapeutic effects. AMG 331 has been shown to have potent inhibition against the human liver enzyme, glycogen synthase kinase-3β (GSK-3β), which plays a role in cancer cell proliferation, inflammatory bowel disease and blood pressure regulation. This drug also has pharmacokinetic properties, meaning that it is absorbed quickly and eliminated slowly from the body, resulting in a long half-life. AMG331 is metabolized by hydrolysis or oxidation by cytochrome P450 enzymes, reduction by glutathione reductase or conjugation with glucuronic acid. Metformin may also inhibit fatty acid synthesis and enhance peripheral glucose uptake in adipocytes.
