TC-E 5008 is a drug that inhibits the proliferation of melanoma tumor cells by blocking the phosphoinositide 3-kinase (PI3K) signaling pathway. TC-E 5008 has been shown to inhibit the growth of cancer cells in vitro and in vivo, with an IC50 of 5 nM and no detectable toxicity. TC-E 5008 has also been shown to be effective against other cancers including leukemia, breast, prostate and colon cancer. The mechanism of action is not known, but TC-E 5008 may function as a competitive inhibitor for phosphatidylinositol 3′ kinase (PI3K). This could lead to the inhibition of downstream targets such as Akt and mTOR, which are key components in cell cycle progression.
