Beschreibung:
Teriparatid ist eine synthetische Form des Parathormons (PTH), die hauptsächlich zur Behandlung von Osteoporose eingesetzt wird. Es zeichnet sich durch seine Fähigkeit aus, die Knochenbildung zu stimulieren, was es von anderen Osteoporosebehandlungen unterscheidet, die sich hauptsächlich auf die Hemmung der Knochenresorption konzentrieren. Teriparatid wird durch subkutane Injektion verabreicht und wird typischerweise für Patienten mit hohem Frakturrisiko verschrieben. Die Substanz besteht aus den ersten 34 Aminosäuren des menschlichen PTH, die für ihre biologische Aktivität entscheidend sind. Ihr Wirkmechanismus besteht darin, an PTH-Rezeptoren auf Osteoblasten zu binden, das Knochenwachstum zu fördern und die Calciumaufnahme im Darm und in den Nieren zu erhöhen. Teriparatid hat eine relativ kurze Halbwertszeit, was eine tägliche Verabreichung für optimale therapeutische Effekte erforderlich macht. Zu den häufigen Nebenwirkungen können Übelkeit, Schwindel und Wadenkrämpfe gehören. Aufgrund seiner anabolen Wirkung auf den Knochen wird Teriparatid häufig für den Einsatz bei postmenopausalen Frauen und Männern mit Osteoporose in Betracht gezogen, die eine Vorgeschichte von Frakturen oder signifikantem Knochenverlust haben.
