Felodipin

Felodipin ist ein Dihydropyridin-Calciumkanalblocker, der hauptsächlich zur Behandlung von Bluthochdruck und Angina pectoris eingesetzt wird. Er wirkt, indem er selektiv das Eindringen von Calciumionen in die glatte Gefäßmuskulatur und das Herzmuskel während der Depolarisation hemmt, was zu einer Vasodilatation und einem gesenkten Blutdruck führt. Felodipin zeichnet sich durch seine lipophile Natur aus, die seine Absorption und Bioverfügbarkeit verbessert. Die Verbindung erscheint typischerweise als gelbes kristallines Feststoff und hat eine relativ lange Halbwertszeit, die eine einmal tägliche Dosierung ermöglicht. Seine Pharmakokinetik wird von Faktoren wie der Nahrungsaufnahme beeinflusst, die seine Absorption beeinträchtigen kann. Felodipin wird in der Leber metabolisiert, hauptsächlich durch Cytochrom-P450-Enzyme, und seine Metaboliten werden hauptsächlich über den Urin ausgeschieden. Zu den häufigen Nebenwirkungen können periphere Ödeme, Kopfschmerzen und Hitzewallungen gehören. Aufgrund seines Wirkmechanismus ist Felodipin wirksam bei der Behandlung von Herz-Kreislauf-Erkrankungen, sollte jedoch bei Patienten mit bestimmten bestehenden Erkrankungen oder bei Patienten, die spezifische Medikamente einnehmen, die mit seinem Metabolismus interagieren können, mit Vorsicht angewendet werden.
