Leflunomid ist ein immunmodulatorisches Medikament, das hauptsächlich zur Behandlung von rheumatoider Arthritis und psoriatischer Arthritis eingesetzt wird. Es funktioniert als Prodrug, was bedeutet, dass es im Körper in seine aktive Form, A77 1726, metabolisiert wird, die das Enzym Dihydroorotat-Dehydrogenase hemmt. Diese Hemmung stört die de novo-Synthese von Pyrimidinen, was zu einer Verringerung der Lymphozytenproliferation und einer Modulation der Immunantworten führt. Leflunomid zeichnet sich durch seine relativ lange Halbwertszeit aus, die eine einmal tägliche Dosierung ermöglicht.
