3-Formyl-2-hydroxybenzonitrile (3FHB) is a potent inhibitor of the E3 ligase that prevents ubiquitin from binding to its target proteins. It is used in cancer therapy as an activator of ubiquitination. 3FHB has been shown to be effective in inhibiting tumour growth and inducing apoptosis in a number of cancer cell lines, including those derived from breast, prostate, brain, and lung cancers. 3FHB also inhibits the proliferation of human colon adenocarcinoma cells by promoting ubiquitination of cyclin D1, which regulates the G1/S transition.
