2,4-Dinitrophenyl β-D-galactoside is a covalent and kinetic inhibitor of β-galactosidase. It is an aglycon that reacts with nucleophiles to form a transition state, which is then converted into the product. The rate of reaction depends on the concentration of nitrophenol, but the enzyme can be inhibited by high concentrations of 2,4-dinitrophenyl β-D-galactoside. The kinetic parameters for this reaction have been determined using isotope effects and are dependent on the concentration of substrate. The mechanism for this reaction involves a nitration step followed by an oxidative deamination step in which the intermediate nitronate reacts with water to form hydroxylamine.
