Histone H2A.Bbd (Barr body deficient) is a variant of the core histone protein H2A, which along with H2B, H3 and H4 form the histone octamer around which DNA is wound to form the nucleosome. Histone variants can replace canonical histones in certain cells or stages of development and help regulate numerous nuclear processes including transcription, DNA repair and chromosome segregation.The genes encoding H2A.Bbd are expressed in a variety of tissues and cell types, suggesting a widespread role in chromatin regulation. H2A.Bbd-containing chromatin has an unconventional chromatin structure which is more open and less stable and is associated with active transcription/ H2A.Bbd-containing nucleosomes arrange only ~120 bp of DNA and have shorter nucleosome spacing. The specific structure of H2A.Bbd bound chromatin facilitates both transcription and initial mRNA processing. H2A.Bbd regulates interactions with RNA splicing machinery and is involved in splicing of introns from precursor mRNAs.
