2-Acetylaminophenylboronicacid (2-APBA) is a postulated prodrug that is activated by dehydration. Dehydration of the 2-APBA molecule results in the formation of a hydrogen bond, which then facilitates nucleophilic attack on the carboxylate. The amide group acts as a leaving group and is replaced by the newly formed carboxylate. This mechanism has been proposed to be an activation step for 2-APBA. Although 2-APBA does not have any acidic properties, it does have a strain energy that is higher than 5-nitrosalicylic acid, which may contribute to its thermal reactivity.
