4-Benzyloxyindole-3-acetonitrile is a calcium antagonist that is used in the treatment of hypertension. It has been shown to inhibit the binding of calcium to cardiac and smooth muscle cells, thereby reducing the contraction rate of these cells. This agent also inhibits the release of noradrenaline and acetylcholine from nerve endings, which may be due to its ability to block the activity of calcium at nerve terminals. This drug binds selectively to the alpha subunit of voltage-gated L-type calcium channels in cardiomyocytes and vascular smooth muscle cells, with an IC50 value of 0.58 μM.
