2,4-Dichlorothieno[3,2-d]pyrimidine is a potent inhibitor of epidermal growth factor receptor (EGFR) tyrosine kinase. It has been shown to inhibit the proliferation of cancer cells in vitro, as well as to reduce tumor size and weight in vivo. 2,4-Dichlorothieno[3,2-d]pyrimidine has potent inhibition against EGFR and is selective for this kinase over other members of the same protein family. Molecular modeling studies have shown that this compound binds to the ATP binding site of EGFR with high affinity and specificity. Cellular assays have shown that 2,4-Dichlorothieno[3,2-d]pyrimidine inhibits cell proliferation by inhibiting EGFR signaling pathways.
