Beschreibung

Donepezil is a cholinesterase inhibitor used in the treatment of Alzheimer's disease. The bioavailability of donepezil is enhanced by the presence of food in the stomach, as well as its metabolism by cytochrome P450 enzymes. Donepezil is metabolized via oxidation and hydroxylation to form 6-O-desmethyl donepezil (6OMD). A pharmacokinetic study was conducted with 6OMD using a whole-cell patch-clamp technique on human erythrocytes. The concentration of 6OMD in plasma was analyzed by LC-MS/MS. This study found that the pharmacokinetics of 6OMD are linear and that it has an elimination half-life of about 15 hours and a volume of distribution at steady state (Vss) of about 500 L. Linear regression analysis showed that there was no significant difference between the mean plasma concentrations for 6OMD and its enantiomer, which suggests that they are
