It is an intermediate in the biosynthesis of maleic acid from succinic acid and it can be synthesized by the addition of potassium ion to malonic acid. 2,2-Dimethylsuccinic acid has been shown to inhibit mitochondrial complex I activity in rat kidney cells. Complex I is responsible for oxidative phosphorylation and the generation of ATP, which provides energy for cell growth and function. This inhibition leads to a decrease in metabolites such as malonic acid and ethylmalonic acid. The accumulation of these metabolites causes metabolic changes that are observed in rats fed with 2,2-dimethylsuccinic acid (e.g., increased levels of polycarboxylic acids).
