2,2-Dimethylsuccinic acid is a chiral compound that has two asymmetric carbon atoms. It is an intermediate in the biosynthesis of maleic acid from succinic acid and it can be synthesized by the addition of potassium ion to malonic acid. 2,2-Dimethylsuccinic acid has been shown to inhibit mitochondrial complex I activity in rat kidney cells. Complex I is responsible for oxidative phosphorylation and the generation of ATP, which provides energy for cell growth and function. This inhibition leads to a decrease in metabolites such as malonic acid and ethylmalonic acid. The accumulation of these metabolites causes metabolic changes that are observed in rats fed with 2,2-dimethylsuccinic acid (e.g., increased levels of polycarboxylic acids).
