Diaminomaleonitrile is a membrane-permeable antimicrobial agent that belongs to the group of nitrogen-containing heterocyclic compounds. It has been reported as an inhibitor of bacterial growth and is used in medical devices such as catheters, urinary drainage tubes, and gloves. The mechanism of action of diaminomaleonitrile involves binding to the enzyme DNA gyrase and inhibiting its activity. Diaminomaleonitrile has been shown to be effective against gram-positive bacteria such as Staphylococcus aureus and gram-negative bacteria such as Escherichia coli. Diaminomaleonitrile binds to the phosphate groups on DNA by steric interactions with the phosphate groups on the enzyme DNA gyrase. This binding prevents the formation of an enzyme-substrate complex required for ATP synthesis, leading to cell death by inhibiting protein synthesis.
