3-Ethynylbenzonitrile is a supramolecular compound that binds to the ATP-binding site of the IκB kinase enzyme and inhibits its activity, which is required for the activation of NF-κB. 3-Ethynylbenzonitrile has been shown to inhibit the production of cytokines associated with inflammation. This compound also inhibits proinflammatory cytokines in mouse models of sepsis, suggesting that it may have therapeutic potential. The lead compound was found to be an inhibitor at a single crystal x-ray level, and it has been proposed as a possible drug target for inflammatory diseases.
