Beschreibung

Topotecan is a dna topoisomerase I inhibitor. It is a prodrug that is converted to its active form, 10-hydroxy-topotecan, by esterases in the body. Topotecan binds to DNA and prevents it from unwinding during replication. This drug also has stereoselectivity for the minor groove of DNA, which makes it more potent than other drugs with similar activity. The main metabolite of topotecan, 10-hydroxy-topotecan, is an inhibitor of topoisomerase II and III. Topotecan has been shown to be more efficient in inhibiting the growth of cancer cells than podophyllotoxin.
