SKF-96365 is a drug that binds to the channel of the ryanodine receptor, which is involved in calcium release and intracellular signaling. It has been shown to be a potent inhibitor of neuronal cell death and myocardial infarct. SKF-96365 was also found to inhibit the polymerase chain reaction by binding to DNA. The binding site for SKF-96365 was identified as being adjacent to the dinucleotide phosphate (dNTP) binding site on one side and near the channel pore on the other. This binding prevents nucleotides from entering or exiting the channel, thereby preventing transcription and replication. SKF-96365 has also been shown to inhibit growth factor-β1 induced calcium entry into HL60 cells, which may contribute to its cardioprotective effects.
