HWY 5069 is an isoquinoline alkaloid that has been found to inhibit the activity of kinases. This inhibitor binds to the ATP binding site of the enzyme, blocking the transfer of phosphate groups and preventing protein phosphorylation. It has been shown to inhibit wild-type protoberberine type I kinase (PK1) and a number of mutants, including PK1B, PK2A, PK2B, PK3A, PK3B and PK4. The IC50 values for these kinases range from 0.5 nM to 7 nM. In addition to its inhibitory effects on kinases, HWY 5069 also inhibits bacterial growth by binding to ribosomes at a site that is different from the binding site for erythromycin or clindamycin.
