Nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that blocks HIV replication by inhibiting the enzyme reverse transcriptase. It has a long plasma half-life and can be taken once daily. Nevirapine is metabolized by cytochrome P450 enzymes in the liver, so impaired liver function can cause decreased clearance of the drug. Nevirapine is well tolerated, but can cause side effects such as nausea, abdominal pain, headache, and rash. Nevirapine is not active against other viruses and it should not be used in patients with hepatitis B or C because of increased risk of hepatic toxicity.
