6-Hydroxypicolinic acid is a hydroxylated derivative of picolinic acid that has been shown to be a potent inhibitor of microbial metabolism. It is an intermediate in the catabolism of picolinic acid, which is formed by the oxidation of tryptophan. The compound can be synthesized from 6-hydroxypyridine and picolinic acid. 6-Hydroxypicolinic acid inhibits bacterial growth by binding to the enzyme nicotinamidase, which catalyzes the final step in cellular nicotinamide adenine dinucleotide (NAD) biosynthesis. This prevents NAD from being converted into nicotinamide adenine dinucleotide phosphate (NADP), thereby disrupting the redox balance within cells.
