Hoechst 33258 is a fluorescent dye that binds to the DNA of cells and can be used as a model system in cell biology. It is also used to detect radiation exposure and has been shown to inhibit the growth of certain bacteria. Hoechst 33258 binds with high affinity to basic proteins, such as histones, which are rich in arginine. This interaction inhibits the ability of DNA polymerase to synthesize DNA and prevents cellular transformation. The drug also inhibits cellular physiology by inhibiting protein synthesis, which may lead to cell death. Hoechst 33258 is not active against most bacteria because it has low affinity for phosphate groups on the bacterial cell wall.
