Nelociguat is a soluble guanylate cyclase stimulator that is used to treat chronic kidney disease. It is the first agent to be approved for this indication in China, and has been shown to improve renal function and reduce proteinuria in Sprague-Dawley rats with chronic kidney disease. Nelociguat stimulates the soluble guanylate cyclase (sGC) enzyme, which increases levels of intracellular guanosine 3',5'-cyclic monophosphate (cGMP). This leads to an increase in nitric oxide production and improved blood flow. Nelociguat has also been shown to have a favourable safety profile and was well tolerated in clinical trials involving healthy Chinese subjects. The drug does not affect male or female fertility, and does not cause changes in haemodynamic parameters such as heart rate, blood pressure, or peripheral vascular resistance.
