5-Nitroorotic acid (5NOA) is a pyrimidine derivative that inhibits protein synthesis. It acts as a competitive inhibitor of the enzyme, orotic aciduria, which converts orotidine 5′-monophosphate to uridine monophosphate. Orotic aciduria is one of the enzymes in the pathway for the synthesis of pyrimidines and purines. This compound has been shown to inhibit the enzyme by binding to its active site and forming a stable complex with it. The inhibition of this enzyme leads to an accumulation of orotic acid and deoxyribonucleotides in cells, causing cellular death.
