• Beschreibung: Pravadoline is a synthetic cannabinoid from the aminoalkylindole family which acts as an agonist at the CB1 receptor (IC50= 453 nM)1. It is also a modulator of prostaglandin synthesis through the inhibition of cyclooxygenase. In vitro experiments on both human and murine breast cancer cell lines showed antiproliferative effects in conjunction with radiation2. Toxicology studies suggested nephrotoxicity in canine models3. In vitro and ex vivo assays showed inhibition of prostaglandin synthesis in mice brain, but unlike NSAIDs, did not provoke gastrointestinal lesions from in vivo oral administration. In animal studies, it showed potent analgesic effects in terms of response latency, prevention of hyperalgesia and anti-nociceptive activity against a range of mechanical, chemical and thermal stimuli1.
