Ractopamine is a non-competitive inhibitor of the enzyme α-ketoglutarate dehydrogenase (KGDH) in the TCA cycle. The KGDH enzyme catalyzes the oxidation of α-ketoglutarate to succinyl coenzyme A, which is an essential step in energy production and amino acid metabolism. Ractopamine has been shown to reduce body fat accumulation in Sprague Dawley rats. It also inhibits the synthesis of ent-kaurane diterpenoids, which are important for controlling inflammation, blood pressure, and cholesterol levels. Ractopamine is used as an additive in livestock feed to promote leanness and increase muscle mass. It increases protein synthesis while decreasing protein degradation by increasing the activity of the ribosomal protein S6 kinase. Spectroscopic data show that rac pterosin B binds with a stereoselective mechanism to its target site on KGDH.
