1,5,9-Triazacyclododecane is a nucleotide that is used in transfection experiments. It can be synthesized from hydrochloric acid and sodium azide in the presence of an organic base such as triethylamine or pyridine. The nucleophilic nature of 1,5,9-triazacyclododecane enables it to form stable complexes with hydrophobic compounds. This property has been shown to be due to the hydrophobic effect. Hydrolysis of these complexes by esterases leads to the release of fatty acids and nucleotides such as ATP. The uptake of 1,5,9-triazacyclododecane into cells has been studied in fetal bovine tissue and also in mammalian cell cultures. These studies have revealed that its uptake is primarily due to passive diffusion with a small contribution from carrier-mediated transport.
