2-Ethoxy-5-nitrobenzoic acid is a potent and selective PTP1B inhibitor. The discovery of 2-ethoxy-5-nitrobenzoic acid was enabled by the use of computational chemistry to design new inhibitors with improved selectivity for PTP1B. Computational studies showed that 2-ethoxy benzoic acid, a known inhibitor of tyrosine phosphatases, binds to the catalytic site of PTP1B. The crystal structure of the complex shows that 2-ethoxy benzoic acid interacts with PTP1B in an orientation very similar to that found in previous studies. Molecular docking experiments show that 2-ethoxy benzoic acid binds to the same region as other known inhibitors and induces conformational changes in the protein. These results suggest that 2-ethoxy benzoic acid may be a good candidate for studying and developing drugs against diabetes and related diseases.
