(R)-(+)-N-Methoxymethyl-n-(trimethylsilyl)methyl-1-phenylethylamine is a potent and selective inhibitor of the protein tyrosine kinase Janus, which has been shown to be involved in myeloproliferative disorders. This drug binds to the active site of Janus and inhibits its kinase activity. The binding of (R)-(+)-N-Methoxymethyl-n-(trimethylsilyl)methyl-1-phenylethylamine to the aralkyl group in Janus leads to a conformational change that prevents ATP from binding. This causes an accumulation of inactive Janus and ultimately cell death. The phenyl group in this drug also acts as a transition state analog for the reaction catalyzed by Janus. The residue at position 3 is important for this drug's inhibitory activity, because it binds with the ligand and stabilizes it
