Glucose 6-phosphate isomerase (GPI) is a key enzyme in the metabolism of glucose. It catalyzes the conversion of glucose-6-phosphate to fructose-6-phosphate, which is an important precursor for glycolysis and gluconeogenesis. GPI has been shown to have a role in chronic arthritis and autoimmune diseases. The compound has been used as a target for antimicrobial peptides and small molecules that inhibit its activity. GPI also plays a role in transcriptional regulation through binding to DNA polymerase, preventing transcription and replication. This enzyme may be involved in energy metabolism, as it regulates the rate of NADH production from NAD+ during glycolysis. GPI is localized to the cytoplasm and nucleus of cells, where it regulates gene expression by controlling ribitol dehydrogenase activity. Disease activity has been correlated with decreased GPI levels in mouse tumor models, suggesting that this enzyme may play a role
