Flagellin binds to the histidine-rich regions of mammalian cells and triggers the production of molecules that activate Toll-like receptor 4 (TLR4) on macrophages, leading to inflammation. Flagellin has also been shown to have immunomodulatory effects, which may be due to its ability to bind insulin receptors and block the insulin signalling pathway. This can lead to insulin resistance and type 2 diabetes. Flagellin has also been shown to inhibit influenza virus replication in vitro and in vivo, as well as stimulate B cell proliferation.
