Beschreibung

2'-Amino-2'-deoxyadenosine (2'ADA) is a nucleoside analogue with potent antiproliferative activity. It inhibits the production of growth factors, such as fibroblast growth factor-β1 (FGF-β1), by binding to divalent metal cation sites in the DNA duplexes, thereby inhibiting transcription and translation. 2'ADA has also been shown to inhibit collagen gel contraction and cyclase activity in vitro. 2'ADA is metabolized in vivo by deamination to form cytidine and 2-deoxyinosine, both of which are biologically active. These metabolites are excreted through glomerular filtration and renal tubular secretion, respectively. The plasma clearance of 2'ADA is approximately 0.3 mL/min per kg body mass index (BMI). This drug binds to human serum albumin, which leads to its long half-life time of about 12 hours.
