NSC 624206 inhibits proteasome activity by binding to the catalytic subunit of the 20S proteasome, which leads to a decrease in ubiquitinated proteins and a reduction of inflammation in macrophages. The clinical response in diabetic patients with neuropathy was evaluated using a cross-over design, and it was found that there was a greater improvement in nerve conduction velocity when compared with placebo.
