The synthesis of 5'-O-levulinoylthymidine 3'-CE phosphoramidite starts with the reaction of 1 equivalent of thymidine with 1 equivalent of levulinic acid in aqueous solution at pH 7 to 8 at 100°C for 2 hours, followed by purification by HPLC.
