2-Hydroxysaclofen is a cholinergic agent that is structurally related to baclofen. It has been shown to have antinociceptive and locomotor-stimulating effects in mice, which may be due to its ability to bind to the 5-HT2 receptors. This drug also blocks the neurotransmitter glutamate at NMDA receptors in the spinal cord and brainstem, which may also contribute to its analgesic effects. 2-Hydroxysaclofen has been shown to produce a dose-dependent sedative effect in rats, with sedation occurring at doses of 0.1 mg/kg or higher. It also inhibits dopamine release from striatal synaptosomes, indicating inhibition of dopamine uptake by presynaptic neurons.
