5'-O-Levulinoylthymidine 3'-CE phosphoramidite is a novel, high quality diphosphate nucleoside. It is modified with levulinic acid and has antiviral and anticancer properties. 5'-O-Levulinoylthymidine 3'-CE phosphoramidite is synthesized from 5'-O-levulinoylthymidine monophosphate, which is synthesized from thymidine and levulinic acid. The synthesis of 5'-O-levulinoylthymidine 3'-CE phosphoramidite starts with the reaction of 1 equivalent of thymidine with 1 equivalent of levulinic acid in aqueous solution at pH 7 to 8 at 100°C for 2 hours, followed by purification by HPLC.
