N-Butyl-4-chloro-3-nitrobenzenesulphonamide is a blocker of voltage-gated sodium channels. It is a competitive antagonist that binds to the voltage sensor region of the channel, where it blocks sodium ion flow and prevents the channel from opening. N-Butyl-4-chloro-3-nitrobenzenesulphonamide has been shown to be pharmacologically active in heterologous systems and inhibits conformation changes in the channel gating machinery. It also binds to channels at concentrations well below those needed for steady state inactivation, leading to time constants that are much longer than those observed for most blockers. N-Butyl-4-chloro-3 nitrobenzenesulphonamide has been shown to block the NaV1.8 sodium channel, which is expressed in peripheral nerve axons and myelinated fibers of the central nervous system, but not in cardiac or smooth muscle cells
