10-Hydroxy imipramine is a metabolite of the antidepressant clomipramine. It is formed by CYP2D6 metabolism and has been found to have similar pharmacological effects as clomipramine, including inhibition of norepinephrine reuptake and inhibition of serotonin uptake. 10-Hydroxy imipramine is a potent inhibitor of human liver microsomal p450 activity and has been shown to be metabolized by microbial enzymes in vitro. This drug also inhibits fatty acid oxidation in rats and may cause infantile hypotonia. 10-Hydroxy imipramine is excreted in breast milk and has a long half-life, which may make it useful for the treatment of depression with insomnia.
