Imidazo[1,5-a]pyrazin-8-amine is a potent and selective inhibitor of the mTORC1 (mammalian target of rapamycin complex 1) kinase. It is orally bioavailable, has high metabolic stability, and a good selectivity profile. Imidazo[1,5-a]pyrazin-8-amine was discovered through an optimization process that yielded molecules with improved potency and selectivity for inhibition of mTORC1. The discovery was made in a xenograft model using human tumor cells lines. Imidazo[1,5-a]pyrazin-8-amine inhibits tumor growth by inhibiting the activity of mTORC1, which plays a key role in the regulation of cell proliferation and cell size.
