2-Iodo-3-methylbenzonitrile is a reactive inhibitor that has been shown to inhibit the catalytic domains of protein kinases. 2-Iodo-3-methylbenzonitrile binds to the ATP binding site and competitively inhibits ATP binding, which prevents phosphorylation of proteins and subsequent activation of the kinase. This drug has been shown to be effective against a number of protein kinases with diverse structures and selectivity profiles. The crystal structure for 2-iodo-3-methylbenzonitrile was determined by X-ray crystallography, revealing a planar molecule with two iodine atoms in close proximity to one another.
