Expressed in E.coli, this is a full length recombinant nucleocapsid protein of the Severe actute respiratory syndrome coronavirus 2 (SARS-CoV-2), containing a 6x His tag at the C-terminal. This product is presented as a frozen liquid in PBS and 25mM K2CO3 and it is important to note that repeated freeze-thaws should be avoided. As an application it may be suitable for ELISA or lateral flow type assays.
First reported in December 2019, SARS-CoV-2 causes Coronavirus Disease-2019 (COVID-19) infections which although can be asymptotic in some individuals it can cause symptoms such as: shortness of breath, fever and in more vulnerable individuals, death. The SARS-CoV-2 nucleocapsid protein is a RNA binding protein responsible for the positive- sense single-stranded RNA genome to be packaged into a helical ribonucleocapsid (NP) complex. Its association with both the viral genome and membrane protein allow it to assist in virion assembly. Additionally studies have shown that the nucleocapsid protein plays a role within the host cells such as interferon inhibition, RNA interference and apoptosis. Structurally it is composed of five domains, three of which are predicted to be disordered: a N-terminal domain, central linkers and a C-terminal domain. Although all five domains are involved with RNA binding, the nucleocapsid protein has an RNA-binding domain and lastly a dimerization domain. Using its disordered regions, protein to protein and protein to RNA interaction sites can occur. Due to the immunodominant nature of the nucleocapsid protein it can function as a diagnostic antigen hence can be clinically used in the diagnosis of SARS-CoV-2.
