Descripción:Pro-opiomelonocortin  (POMC) is cleaved into many products including β-Lipotropin.  β-Lipotropin is a precursor of several peptides including β- melanocyte-stimulating hormone (β-MSH). β-MSH is primarily found in the pituitary gland. β-MSH regulates feeding and levels change with the nutritional status. β-MSH is a melanocortin receptor agonist, particularly MC4R. The hypothalamic melanocortin system provides the most robust neural signals to inhibit caloric intake. A better understanding may provide novel methods of manipulation to tackle obesity.β-MSH directly stimulates the corticotropic axis and cortisol inhibits β-MSH. β-MSH's role is to optimize the general catabolic activity of the organism in an immediate manner. β-MSH stimulates cortisol release without the aid of adrenocorticotropic hormone (ACTH) and pulls thyroid activity outside the vertical feedback information of thyroid releasing hormone (TRH) and thyroid stimulating hormone (TSH).β-MSH(1-18) levels have been shown to significantly reduce following cells' exposure to fatty acid palmitate. LC-MS/MS is effective at detecting POMC-derived peptides and shows fatty acids can dramatically reduce the production of POMC-derived peptides including β-MSH(1-18). Hypothalamic inflammation triggered by dietary fats can affect POMC neurons. The work with β-MSH(1-18)  helps provide insight into the mechanism behind diet-induced obesity.This β-MSH(1-18) phenylalanine residue is isotopically labelled with carbon-13 (9) and nitrogen-15 (1), giving this peptide a mass increase of 10 compared to the unlabelled peptide. This will allow absolute quantification applications such as quantitative proteomics, the quantification of complex protein mixture at very low concentrations, or NMR studies.
