3-Bromothiophene-2-carbonitrile is a reactive electrophile that serves as an intermediate for the synthesis of heterocycles. It is prepared by reaction with triethyl orthoformate and methoxy in the presence of sulfur, followed by thermolysis. The structure activity relationship of this compound has been studied using decahydroisoquinoline and methoxy group as models. Electron distribution in the nucleus and electron affinity enhancements are observed when 3-bromothiophene-2-carbonitrile is conjugated to azides or distal groups.
