(S)-tert-Butyl 3-hydroxybutanoate is a chiral substrate for the production of optically pure (R)-3-hydroxybutanoic acid. The reaction can be catalyzed by the fungus Rhizopus arrhizus and the model substrate used in this study was acetophenone. The reaction proceeds with high selectivity, giving a yield of 72% and an enantiomeric excess of 98%. The optimal conditions were determined to be at pH 7, a substrate concentration of 2.5 mmol/L, and a reaction time of 2 hours. These results have implications for laboratory syntheses that require the use of chiral substrates with high selectivities and yields.
