Bis-(1,3-dibutylbarbituric acid)pentamethineoxonol is a potent inhibitor of the proton pump. It has been shown to inhibit the activity of the proton pump in rat stomachs and also in human gastric parietal cells. The inhibition is reversible, and bis-(1,3-dibutylbarbituric acid)pentamethineoxonol does not alter the resting membrane potential or ion distribution. Bis-(1,3-dibutylbarbituric acid)pentamethineoxonol was found to be nonlethal for cultured human lung cells and was shown to inhibit the growth of HL60 cells with an IC50 value of 0.4 µM. This drug binds to a specific site on the enzyme H+/K+ ATPase, which is located on the outer surface of the plasma membrane.
