4-Chlorothieno[3,2-c]pyridine (4C) is an inhibitor of human phosphoribosyltransferase that belongs to the class of allosteric inhibitors. 4C has high potency and efficiency in assays and is an efficient inhibitor of phosphoribosyltransferase. It binds to a site on the enzyme that is close to the active site, which prevents substrate binding. 4C has been shown to inhibit cellular function by inhibiting protein synthesis and DNA replication. The mechanism of inhibition involves binding to a site on the enzyme's β subunit that is near the active site and preventing substrate binding. 4C also inhibits ATP hydrolysis at low concentrations, but it does not inhibit ATP formation at any concentration tested.
