2-Chloro-6-hydroxynicotinic acid is a nicotinamide analog that inhibits the function of nicotinic acid receptors in animals. It is hydrolyzed to 2-chloronicotinic acid, which inhibits the enzyme phosphotransferase and prevents the release of acetylcholine from presynaptic nerve terminals. The hydrolysis of 2-chloro-6-hydroxynicotinic acid also leads to an increase in cellular lipids and lipid peroxidation, which may contribute to its antiinflammatory effects. Hydrolysis by esterases or glucuronidases results in an active form that can be oxidized by cytochrome P450 enzymes, reduced by glutathione reductase, or conjugated with glucuronic acid.
